Semen of 20 Holstein-Fresian bulls aged 3-5 years were collected and frozen once a week for 3 weeks. The result showed the post-thaw sperm motility and progressive motility at 0 h from the 87% and 99% glycerol were not significantly different (P>0.05). The ALH (amplitude of lateral head displacement) and BCF (beat cross frequency) of the sperm from the extender using the 99% glycerol was significantly higher (P<0.05). However, after 5 h of incubation at 37?C the 87% glycerol gave significantly lower motility and progressive motiloty with higher VAP (average path velocity), VCL (curvilinear velocity) and ALH (P<0.05). The results of dilution methods did not differ significantly at 0 h. except at 5 h. the one-step method revealed significant higher motility (P<0.05. There were no effects of glycerol cencentration and dilution methods on post-thaw sperm head elongation, area, plasma membrane and acrosomal integrity (P>0.05). In conclusion, using 99% glycerol and one-step dilution methods gave a better semen quality. |

This study investigated the usefulness of artificial insemination in the Thai native crossbred horse breed in order to increase reproductive success and improve breeding management and the genetics of the equine population in Thailand. Semen, collected from a 7-year-old Thai native crossbred stallion was frozen using the liquid nitrogen vapor freezing method. An insemination dose of 50 ? 106 motile cells frozen semen was used in one intrauterine artificial insemination and resulted in the pregnancy and birth of the first healthy Thai native crossbred offspring. |

In the first experiment, semen samples were diluted 1:1 (v:v) with two different semen extenders, Beltsville poultry semen extender (BPSE) or modified Tyrode’s medium (TALP) and cryopreserved with either 6% dimethyl acetamide (DMA) or 8% dimethyl sulfoxide (DMSO). The best result (p<0.05) was obtained for TALP extender + 8% DMSO, with values of 57.32% for viability, 56.57% for normal morphology and 48.50% for motility. In experiment 2, two different freezing protocols (one-step and two-step) were evaluated. The results showed that there was no difference in post-thaw motility and viability of the spermatozoa (sperm) when either the one-step or the two-step protocol was applied with 8% DMSO. In contrast, the sperm motility and viability with the one-step protocol were better (p<0.05) than those with the two- step protocol when 6% DMA was used. This study indicated that from in vitro analysis, TALP with 8% DMSO was the preferred freezing solution for three-yellow cock sperm and this could be carried out using a one-step freezing protocol. |
